Can I run an online game via email, Skype, Google Hangout or a play-by-post forum?
Yes, you may run games via Skype, Google Hangout or play-by-post forum provided you do not post or upload any transcripts from your playtest, upload any playtest materials and make sure that all members of your group have signed up to playtest through the official playtest signup process.
You would not be posting or uploading a transcript from a playtest you were running so yes you could post a transcript of it.

Can we run this playtest at home or is it only in stores, like D&D Encounters or D&D Lair Assault?

You are welcome to run the D&D Next playtest at home. However, all members of the group must follow the official sign-up process at DNDNext.com and agree to the terms and conditions for playtesting.
And don't think you'll get away with it if your second cousin Larry happens to stop by on the day and decides to take part without signing up. Wizards will know. Wizards always know.

If your second cousin Larry happened to stop by on the day and decided to take part without signing up then you'd be breaking the OPTA and that would be very bad.

I know I shouldn't be looking a gift horse in the mouth (if that anecdote even applies here, I'm not sure) but I'm not sure what it means to post a transcript of a PBP game. I mean... isn't it already a transcript? ...that is posted?

edit: Nevermind. Found my question answered(ish) in another thread. Still seems silly to me.

Lune - It is worded very strangely. I made the same point at ENWorld and read a similar series of posts at WotC. The general consensus appears to be that because they are individual posts, it's not a big deal. The concern (which is weird) is if you took every post and put it in one document and then uploaded that.
